Go do WvW and kill people ... I mean, it's very straightforward. You can easily hit 50 in an hour or two of play.

If you're having a difficult time killing people running around solo, try to find a decently sized zerg and follow them around. You should be able to kill enough people in a reasonable time then.
Being in a zerg is the only thing that already works for me. Still far away on the 50, Im in 36.
Some people says tome that I need to use ballistas and something like that.
I've been WvW for a while and have about 7800 kills, it would be higher had I not stopped playing or if my play style wasnt a "roamer". (Half of my kills are from solo fights so that's the VERY SLOW way to get them.)
He's is what is KEY...tagging and/or siege. If you want kills, and you are not in a keep or around siege, make sure you do 20% or more damage to as many people as possible. So rather than focusing on one guy and killing him completely (1 kill) do AOE attacks to 5 peopel that do at least 20% damage (and damage while they;re in downed state counts I believe) and you get 5 kills. You just became 5 times more efficient in getting kills.
The other one is obv, get on an arrow cart and shoot people for huge AOE damage (so you've tagged them) and bigger radius so you tag more people (no aoe cap)...so kills gallore if you're defending a keep or a supply camp with siege and a few people to help so you can actually kill them all.
Boom, achievement done in a hour.

If you can get on arrow cart on defense of a tower or keep you can rack up tons of kills pretty quick. If you are out in the world run with a group, doesn't have to be a zerg, but going solo, unless you are pretty skilled, is a good way to die. And yes AOE. If its a full blown AEO great, if all you have are bounce attacks those will at least help you tag more people to get credit for more kills. My 2x dagger / 2x pistols thief doesn't have much in the way of AOEs other than bounce and the elite skill dagger storm, but unleashing that in the middle of a zerg and then stealing to a warrior and using axe whirl, well i get a kill from just about everyone that dies in the zerg. So if you have spamable aoes you should be able to get your 50 np.
I've got to second what Bokiz said. Tower defense is the easiest way to rack up kills, but being offensive can work too if you're smart with it.
My necromancer regularly gets 50+ kills in an outing by diving head first into zergs to AoE them and debuff them down. A couple of days ago a friend and I completely turned around a fight where our people were outnumbered 3 to 1 by jumping in and alternating our wells and Plague forms. We got 175 kills between the two of us in an hour and eventually their people started targeting us specifically, which only got us more kills when we took down their hit squads. Mesmers also have a great anti-zerg AoE that most people don't even think about in Feedback. If you drop that on a zerg you'll tag pretty much everyone using a ranged weapon. If you coordinate with others you can stack their abilities under a rain of arrows from a cart you can melt almost any zerg. The problem with that is the communication, people just aren't good at it.
My biggest tips for being successful in WvW? Practice, stay calm, and don't be afraid to die. Find a spec that suits your play style and works well for you and use it. Don't listen to people who say there's only one viable spec for whatever class. Don't use the cookie cutter builds that are floating around, or if you do only do it while you're still learning the character. People expect certain builds when they see certain classes. If you show up with something completely different that works you'll blow their minds and cause them panic, and their panic means more kills for you.
