Welcome to the forums and welcome to the Guild Wars franchise! Guild is a pretty common term in the MMO genre and what it basically refers to is an in game group that you can consistently play with and talk to. It's like a ready made social club for you to walk into.
Guilds will usually offer multiple benefits like events, a constant group to run dungeons with, forums specific to the guild, and a VOIP server of some sort so you can all talk via voice rather than text. To be honest, guilds will work a bit different in GW2. Most games only allow you to join one specific guild at any time on a character. You will then represent that guild with the guild name over floating over your head with your character name. In GW2 though you can join many guilds and choose which one you want to represent at any time. Another benefit in GW2 to joining a guild is that by completing actions in the world you gain "influence" for the guild that you a representing. Guild leaders can use that influence to buy buffs, guild storage, and other things for the members.
Besides a social group though, guilds offer in game services as well like being able to help newer players or helping players through harder runs, or even providing them with crafting materials.. You can't be every craft in the game so having a guild full of people with different craftmanship specialties allows you access to a broader range of crafted materials.
So to sum it up...guilds are a lot. They're a social group, a resource for help, materials, gold, buffs, and advice. They are the people you rely on. They're the folks you hang out with regularly.

Great write-up by Murder, and I'll just add a couple of points. 1.) Guilds are also ways of finding specific people who like to play a game like you do. Which is why you'll see guilds claim to be "heavily PVP" or "heavily PVE". Some guilds are into roleplaying, so if you are into that you can find guilds who offer a strong RP'ing presence. 2.) They also offer a way to stay ahead of competition and others in a game. Just ask anyone if a person is considered more of a power/influence if they play solely by themselves or with a guild.

"A guild is a close group of players coming together for the purpose of having a good time. That good time can be achieved by domination of players or the environment or completing goals. Others are just chilling with swag and knocking out monsters. But the point is ultimately to enjoy yourself, and help the other members have the best guild experience EVER."

Although GW2 supposedly will be soloable to complete your story line things like that really are the basics, completing the game. Completing a game is often just a case of unlocking the areas, skills and equipment. Thats when the fun can start to begin and where being in a good guild comes in. With a good guild theres no logging in thinking ok what shall i do today. As you progress you'll get all kinds of help with the idea that you will follow suit when the next generation of new guys comes through. you don't have to complete a title or dungeon to get satisfaction or fun out of a game. A guild is the difference between a RPG and a MMORPG. I have about 14k+ hours in GW1, thats the level of entertainment you can get from a guild, without them and the long wait for GW2 theres no way i would have so many hours, its a good game but no game on its own is that good. If your new to guilds i would suggest looking for one that is about fun and helping one another rather than be too serious. The majority of the serious guilds will come and go and there's plenty of time to get into the good stuff, sit back and enjoy the game with some like minded chilled out friends. Worth noting key to any guild is activity, its dead without it. Guild Wars and Guild Wars 2 have very good quality wiki's so be sure to make use of it, will save you some of those embarassing newb questions. http://wiki.guildwars2.com/wiki/Main_Page
btw fyi we generally have two different meanings for n00b/newb
newb - name given to a new player looking to seek out information and learn the game (everyone will be a newb in GW2)
n00b - a condescending name given to a new player trying to act like they know it all and failing
